Studied at St Andrews and Leiden with period of study in Antwerp (1725). Received MD from University of St Andrew in 1729.
Moved to London and tutored sons of the duke of Rutland at Rutland House, Cavendish Sq, London. By 1746 Clephane was practicing medicine in London and through connection with Lord Sandwhich was appointed physician to the army. Brief stint as superintendent of the army hospital at Ipswich (1749). Clephane returned to London in 1751 as physician at St George’s Hospital, moving to Golden Square. He became a licentiate of the Royal College of Physicians in 1752 and was an original member of the Society of London Physicians. Clephane published articles in its Medical Observations and Inquiries on the successful treatment of lockjaw and the history of gout powder. He was recalled to military service to force at Isle of Wright 1758 but died of illness the same year.
